Pluripotent cell separation using “cell rolling column”
Stem cells are promising cell source, which have been used in a wide variety of fields--e.g. drug developments, medical applications and cell biology. To obtain these cells, cell isolation procedures are necessary because they rarely exists in a much larger population of background cells in our body. We developed a new cell separation device called "cell rolling column", in which, cells selectively roll on antibody-immobilized column surfaces1. Mahara et al. isolated two distinctive cell populations with osteoblastic differentiation ability from bone marrow mesenchymal stem cells2.
In conventional antibody-based cell separation strategies, obtained cells are labeled with affinitive biomolecules, which can adversely affect cell phenotype. In cell rolling column, cell are separated without labeling agents, and cell rolling column can be a promising tool to separate pluripotent cells (including ES or iPS cells).
